Axel started playing football at a young age. In 1994, he began attending the Germinal Ekeren youth school, where he remained until 1999. However, the young people’s school’s name was altered later, and it is now known as Beerschot.
On February 22, 2007, a young Belgian defender agreed to a three-year deal with Ajax, which was his first long-term contract. For the 2008–09 campaign, he was promoted to the first team, although he made his debut on January 18 in a 4–2 win over NEC.

On February 26, Alderweireld made his debut in Europe by drawing 1-1 with Fiorentina in the UEFA Cup. On September 1, 2014, Alderweireld was loaned to Southampton of the English Premier League for one season with the possibility of a purchase at the end of the transfer window. At the end of the season, Southampton only had a £6.8 million incentive to keep Alderweireld, and Atlético removed the provision for the highest bidder in July 2015.
